چکیده

Haemophilm ducreyi has been described as a fastidious organism requiring blood for growth (Greenblatt, 1953). It has been successfully cultivated on clotted blood (Teague and Diebert, 1920) and on various artificial media enriched with blood, (Sanderson and Greenblatt, 1937; Beeson and Heyman, 1945; Beeson, 1946; Deacon et al., 1956). However, little is known concerning the specific nutritional requirements of this organism. In an effort to define the factors necessary for the growth of H. ducreyi this preliminary investigation of a representative virulent strain was begun.

Cytopathic effect of Haemophilus ducreyi for human foreskin cell culture.

An explant adult foreskin cell culture (FS2-3) was compared with human lung carcinoma cell culture (A549) with regard to the ability of Haemophilus ducreyi to produce a cytopathic effect. The survival of H. ducreyi for up to 26 days in FS2-3 cells was far greater than in any previously described in-vitro culture system. H. ducreyi survived for up to 7 days in A549 cells. The H. ducreyi cells gr...

Virulence factors of Haemophilus ducreyi.

We investigated the susceptibility of virulent and avirulent strains of Haemophilus ducreyi to the bactericidal activity of normal human serum and to phagocytosis and killing by human polymorphonuclear leukocytes (PMNL). Strains were defined as virulent if intradermal inoculation into a rabbit produced a typical necrotic lesion. Nonvirulent strains produced no cutaneous lesions in rabbits. Viru...

Involvement of the Haemophilus ducreyi gmhA gene product in lipooligosaccharide expression and virulence.

The lipooligosaccharide (LOS) present in the outer membrane of Haemophilus ducreyi is likely a virulence factor for this sexually transmitted pathogen. An open reading frame in H. ducreyi 35000 was found to encode a predicted protein that had 87% identity with the protein product of the gmhA (isn) gene of Haemophilus influenzae. In H. influenzae type b, inactivation of the gmhA gene caused the ...

Mutations in the lspA1 and lspA2 genes of Haemophilus ducreyi affect the virulence of this pathogen in an animal model system.

Haemophilus ducreyi 35000HP contains two genes, lspA1 and lspA2, whose predicted protein products have molecular weights of 456,000 and 543,000, respectively (C. K. Ward, S. R. Lumbley, J. L. Latimer, L. D. Cope, and E. J. Hansen, J. Bacteriol. 180:6013-6022, 1998). We have constructed three H. ducreyi 35000HP mutants containing antibiotic resistance cartridges in one or both of the lspA1 and l...

Studies on Haemophilus Influenzae

1. It has been shown that the ability of virulent S. influenzae to kill mice is enhanced by suspending the organism in mucin. 2. The occurrence of a true infection characterized by in vivo multiplication of the organism has been established. 3. The necessity for using a pure inbred strain of mice for this type of study has been confirmed. 4. The increase in mucin virulence of a strain of H. inf...
